Whole-genome sequencing reveals host factors underlying critical COVID-19

Whole-genome sequencing, transcriptome-wide association and fine-mapping analyses in over 7,000 individuals with critical COVID-19 are used to identify 16 independent variants that are associated with severe illness in COVID-19.

Tyrosine dephosphorylated cortactin downregulates contractility at the epithelial zonula adherens through SRGAP1

Epithelial cell-cell adhesions are contractile junctions, but whether contractility can be down-regulated is not known. Here the authors report how tyrosine dephosphorylation of the cytoskeletal scaffold, cortactin, recruits the RhoA antagonist SRGAP1 to relax adherens junctions in response to HGF.

Centralspindlin and α-catenin regulate Rho signalling at the epithelial zonula adherens

Yap and colleagues identify an extramitotic role for the centralspindlin complex, a Rho signalling regulator during cytokinesis. They show that centralspindlin interacts with α-catenin, and promotes epithelial junctional integrity by recruiting the ECT2 RhoGEF to the zonula adherens, as well as by inhibiting the junctional localization of the p190 B RhoGAP.

N-WASP regulates the epithelial junctional actin cytoskeleton through a non-canonical post-nucleation pathway

N-WASP activates Arp2/3-mediated actin nucleation. N-WASP is now shown to stabilize and organize the actin cytoskeleton at cell–cell junctions through its interaction with the actin-binding protein WIRE.

Cortical F-actin stabilization generates apical–lateral patterns of junctional contractility that integrate cells into epithelia

Yap and colleagues demonstrate that E-cadherin-based cell–cell junctions exhibit distinct patterns of apical and lateral contractility. They show that N-WASP-dependent stabilization of F-actin mediates increased apical junctional tension, and that modulation of intra-junctional tension differences can promote extrusion of cells from monolayers.

Vaccination is effective in protecting from COVID-19. Here the authors report immune responses and breakthrough infections in twice-vaccinated patients receiving anti-TNF treatments for inflammatory bowel disease, and find dampened vaccine responses that implicate the need of adapted vaccination schedules for these patients.
