Phage Paride can kill dormant, antibiotic-tolerant cells of Pseudomonas aeruginosa by direct lytic replication

Bacteriophages have primarily been studied using fast-growing laboratory cultures of their bacterial hosts. Here, Maffei et al. show that diverse phages can infect deep-dormant bacteria and suspend their replication until the host resuscitates; however, a particular phage can directly replicate and induce the lysis of deep-dormant hosts.

Cyclic di-GMP acts as a cell cycle oscillator to drive chromosome replication

In Caulobacter crescentus, oscillating levels of the second messenger cyclic-di-GMP drive the cell cycle through regulation of the essential cell cycle kinase CckA; as its levels increase during the G1–S transition, cyclic-di-GMP binds to CckA to inhibit kinase and stimulate phosphatase activity, thereby enabling replication initiation.

A deterministic, c-di-GMP-dependent program ensures the generation of phenotypically similar, symmetric daughter cells during cytokinesis

Phenotypic heterogeneity in bacteria can be generated in a programmed manner, by the asymmetrical distribution of c-di-GMP metabolizing enzymes during bacterial cell division. Here, the authors identify a c-di-GMP-dependent program with an opposite effect: the deployment and transient activation of a diguanylate cyclase at the cell division site ensures the formation of phenotypically similar daughter cells in Myxococcus xanthus.

Bacteria adapt their growth rate to their metabolic status and environmental conditions by modulating the length of their G1 period. Here the authors show that an increase in c-di-GMP concentration modulates the activity of kinase ShkA and transcription factor TacA, thus enabling G1/S transition in Caulobacter.

Hartl et al. demonstrate how metabolism and the cell cycle are coupled in prokaryotic cells by analysing the metabolome during the cell cycle in Caulobacter crescentus.
