Two-stage binding of mitochondrial ferredoxin-2 to the core iron-sulfur cluster assembly complex

De novo [2Fe-2S] cluster synthesis by the mitochondrial ISC complex requires electrons from ferredoxin-2 (FDX2). The authors map FDX2 binding to the core ISC complex, showing that FDX2 can bind in two possible conformations in a manner competitive with frataxin.

Functional spectrum and specificity of mitochondrial ferredoxins FDX1 and FDX2

Mitochondrial ferredoxins FDX1 and FDX2 are assigned to specifically donate electrons to steroidogenesis, Fe–S protein biogenesis, heme a formation or lipoylation. The proteins’ functions can be swapped by mutually exchanging short peptide segments.

[2Fe-2S] protein cofactors are essential for life and are synthesized on ISCU2 scaffolds. Here, the authors show that hydrophobic interaction of two conserved N-terminal tyrosines induces ISCU2 dimerization and concomitant [2Fe-2S] cluster synthesis.
