Msx1⁺ stem cells recruited by bioactive tissue engineering graft for bone regeneration

Critical-sized bone defects still present clinical challenges. Here the authors show that transplantation of neurotrophic supplement-incorporated hydrogel grafts promote full-thickness regeneration of the calvarium and perform scRNA-seq to reveal contributing stem/progenitor cells, notably a resident Msx1+ skeletal stem cell population.

Divide-and-conquer strategy with engineered ossification center organoids for rapid bone healing through developmental cell recruitment

Critical-sized bone defects are a clinical challenge, with long-term recovery often leading to delayed union or nonunion. Here, Zhang et al. report an engineered ossification center-like organoid which recruits Krt8+ skeletal stem cells and reduces Has1+ fibrotic cells, mimicking developmental bone formation for regeneration of critical-sized bone defects.

Physical and chemical niche of human growth plate for polarized bone development

Here the authors show that the growth plate (GP)-epiphysis interface possesses a strong modulus transition and acts as a mineralization inhibition zone, while the GP-metaphysis interface has a gradual transition and stimulates bone formation. They also identify proteins at these boundaries which inhibit or promote bone mineralization.

Ebola virus VP35 hijacks the PKA-CREB1 pathway for replication and pathogenesis by AKIP1 association

Ebola virus virion protein 35 (VP35) is a cofactor of the viral RNA-dependent RNA polymerase, required for viral assembly and IFN antagonist. Here, Zhu et al. provide evidence that EBOV VP35 induces an AKIP1-mediated (human A kinase interacting protein) activation of the PKA-CREB1 signaling pathway and contributes to viral replication and pathogenesis in vitro and in vivo.
