Loss-of-function mutations in Keratin 32 gene disrupt skin immune homeostasis in pityriasis rubra pilaris

Pityriasis rubra pilaris (PRP) is a chronic immune‐mediated papulosquamous dermatosis and is caused by dysregulation of keratinocytes. Here the authors identify keratin 32 (KRT32) mutations associated with PRP which results in the inhibitory function of KRT32 towards NF-kB signalling and IKK complex formation being reduced.

A large-scale genome-wide association and meta-analysis identified four novel susceptibility loci for leprosy

Previous studies have shown genetic associations between leprosy and 18 different genes/loci. Here, Wang and colleagues perform genome-wide association study in Han Chinese leprosy patients and describe four novel loci to be associated to the disease.

Furen Zhang and colleagues report the results of a three-stage genome-wide association study of leprosy in the Chinese population. They discover six new susceptibility loci and observe substantial overlap with loci previously implicated in autoimmune and inflammatory diseases.
