Stress-induced RNA–chromatin interactions promote endothelial dysfunction

Global interaction of chromatin-associated RNAs and DNA can be identified in situ. Here the authors report the genome-wide increase of interchromosomal RNA-DNA interactions and demonstrate the importance of such RNA-DNA contacts exemplified by LINC00607 RNA and SERPINE1 gene’s super enhancer in dysfunctional endothelial cell models.

Mapping RNA–chromatin interactions by sequencing with iMARGI

iMARGI (in situ mapping of RNA–genome interactome) is a proximity ligation method for global profiling of chromatin-associated RNAs. A linker sequence bridges DNA and RNA in physical proximity, permitting sequencing library preparation and mapping of DNA–RNA contacts.

miR-379 deletion ameliorates features of diabetic kidney disease by enhancing adaptive mitophagy via FIS1

Kato, Abdollahi et al. identify a redox protein thioredoxin and mitochondrial fission-1 (FIS1) protein as miR-379 targets in mouse kidney. They find that miR-379 knockout mice are protected from diabetic kidney disease by enhancing mitophagy via FIS1, suggesting miR-379 as a potential therapeutic target for diabetic kidney disease.

Downregulation of hepatic lncRNA Gm19619 improves gluconeogenesis and lipogenesis following vertical sleeve gastrectomy in mice

A hepatic lncRNA Gm19619, which is repressed by vertical sleeve gastrectomy but upregulated by obesity and fasting, regulates gluconeogenesis and lipid metabolism in mice through Foxo1-G6pc/Pck1 and Lepr signaling pathway respectively.
