a, Whole cell STED images of Rho-ZF5.3 and LAMP1-SiR show the sub localization of Rho-ZF5.3 within the lysosomal lumen. b, Similarly, whole cell STED images of Rho-BBA5.3 and LAMP1-SiR depict a similar sub-organellar localization. Regions of interest across 6 different cells (3 for Rho-ZF5.3 and 3 for Rho-BBA5.3) were imaged at a higher spatial sampling rate to determine differences in sub-organellar organization of c, Rho-ZF5.3 and d, Rho-BBA5.3. Line profiles across vesicles of interest were plotted. An assortment of phenotypes is seen for both Rho-ZF5.3 and Rho-BBA5.3. Scale bar 5 µm (a, b), and 1 µm (c, d). Images are representative of two biological replicates per condition.
