The effect of Us3 on the recruitment of UL31 to sites of DNA damage. (A) HeLa cells co-expressing EGFP-UL31 with either WT Us3 or kinase dead (KD) Us3 were subjected to laser microirradiation. Images are from immediately before (Pre), immediately after (Post) and 30 seconds after microirradiation. Arrows denote the location of nucleoli. (B) Δmax values from microirradiated cells expressing EGFP-UL31, NBS1 and H2A1.1 macro domain in the presence of WT or KD Us3. Δmax was calculated from n ≥ 14 cells for each co-transfection condition from two independent experiments. Error bars represent standard error of the mean. Using an unpaired t test the Δmax of KD Us3 co-expressing cells was compared to cells co-expressing WT Us3 (**p < 0.01, ***p < 0.001).
