UL31 recruitment to sites of laser microirradiation requires PARP1/2 activity. HeLa 0.1% DMSO (vehicle) or 10 nM olaparib, a PARP1/2 inhibitor, one hour prior to imaging. Δmax values of 0.1% DMSO (−) and 10 nM olaparib treated (+) cells expressing the indicated proteins was determined as described above in Fig. 5. n ≥ 6 cells/experiment in two to four independent experiments for each construct analyzed. Error bars represent standard error of the mean. Using an unpaired t test the Δmax of DMSO treated cells was compared to olaparib treated cells (ns = not significant, **p < 0.01, ***p < 0.001).
