Two distinct Mon/Mac (Ki67+ and TNFα+) subsets increased in IL-23 inflamed skin. (a) Representative 2-dimentional couture plot of TNFα and Ki67 expression in Mon/Mac cells from IL-23 injected ears at day 4 determined by flow cytometry. The quad gate that separates TNFα and Ki67 positive/negative was determined based on comparing fluorescent staining from IL-23 ears to those from naive ears and IL-23 injected ears without the specific fluorescent antibody. Numbers indicate the frequencies of the cell population within the quad gates. (b) Enumeration of Ki67+ and TNFα+ Mon/Mac cells in ears of sham and IL-23 injected animals (n = 6–8).
