Depression of tryptophan may contribute to adverse effects of naproxen

Non-Steroidal Anti-Inflammatory Drugs (NSAIDs) have been associated with adverse effects affecting both the cardiovascular and gastrointestinal systems. Here the authors show that NSAIDs lower plasma tryptophan concentrations in humans and mice and that replacement of tryptophan in mice treated with naproxen leads to decreased adverse effects.

Artificial intelligence guided discovery of a barrier-protective therapy in inflammatory bowel disease

SPT6 promotes epidermal differentiation and blockade of an intestinal-like phenotype through control of transcriptional elongation

The role of RNA polymerase II (Pol II) recruitment during adult stem cell differentiation is well understood, but not that of Pol II elongation. Here, the authors show that 30% of epidermal stem cell differentiation genes depend on SPT6 and PAF1 for Pol II elongation, and SPT6 loss leads to an intestine-like phenotype.

Here, the authors generated a genome-wide map of the global targets bound by HNF4A and HNF1A in beta cells and hepatic cells, and highlighted notable downstream pathways and target genes that may influence beta cell function.
