Cell state as a design variable in nanomedicine

Nanomedicine has advanced through increasingly sophisticated particle design, yet generalizable rules for productive delivery remain scarce. This gap persists in part because the receiving cell is still treated as assay background rather than as an active design variable. Bringing cell state into the design space can help to translate context-dependent performance into more actionable and predictive rules for nanomedicine development.

Deciphering cell types by integrating scATAC-seq data with genome sequences

This study introduces SANGO, a method for accurate single-cell annotation leveraging genomic sequences around accessibility peaks within single-cell ATAC sequencing data. SANGO consistently outperforms existing methods across diverse datasets for identification of cell type and detection of unknown tumor cells. SANGO enables the discovery of cell-type-specific functional insights through expression enrichment, cis-regulatory chromatin interactions and motif enrichment analyses.

Ultra-high-voltage Ni-rich layered cathodes in practical Li metal batteries enabled by a sulfonamide-based electrolyte

Charging at high voltages in principle makes batteries energy dense, but this is often achieved at the cost of the cycling stability. Here the authors design a sulfonamide-based electrolyte to enable a Li metal battery with a state-of-the-art cathode at an ultra-high voltage of 4.7 V while maintaining cyclability.

Critical issues such as oxygen release during battery cycling plague the development of high-energy Li-rich oxide cathodes. Functional tissue-engineered bone-like graft made of a fibrin scaffold and TG2 gene-modified EMSCs for bone defect repair

In this study, ectomesenchymal stem cells (EMSCs) were modified with the transglutaminase 2 (TG2) genes and tested for their ability to enhance bone regeneration on fibrin scaffold. In vitro analyses revealed that osteogenic differentiation of the EMSCs on the fibrin scaffold was promoted by TG2 expression. Transplanting fibrin scaffold loaded with TG2 gene-modified EMSC into bone defects on rat skull induced significantly faster bone regeneration compared with non-genetically modified EMSC.
