Anthracyclines, such as doxorubicin and mitoxantrone, are among the most effective chemotherapies for cancer; they work by interfering with DNA replication and inducing cell death. A new study in Nature Medicine shows that the efficacy of these drugs is also linked to their ability to stimulate a type I interferon (IFN) response through Toll-like receptor 3 (TLR3), an intracellular immune receptor that is involved in sensing viruses.
