Ubiquitylation determines cell fate

The authors analysed the transcriptomes of differentiating human embryonic stem cells (ES cells) and found a strong reduction of KBTBD8 expression during neural differentiation. When KBTBD8-depleted ES cells were induced to undergo neural conversion (that is, to differentiate into neural cells), they failed to produce neural crest cells and instead produced more CNS precursors, particularly those with forebrain identity. Similarly, in Xenopus tropicalis embryos, reduced expression or activity of CUL3KBTBD8 prevented neural crest formation and expanded the territory occupied by CNS precursors. Thus, CUL3KBTBD8 promotes the differentiation of ES cells into neural crest cells by suppressing their differentiation into CNS cells.
