The androgen receptor—a novel target for vaccines

The androgen receptor is a potential target for prostate cancer vaccines, according to a research group from Wisconsin, USA. Olson et al. used several rodent models to demonstrate the efficacy of a DNA vaccine (pTVG-AR) that targets the androgen receptor ligand-binding domain, which demonstrates high levels of intraspecies and interspecies sequence homology.

The researchers found that pTVG-AR immunization of HHDII-DR1 mice—which express human histocompatibility leukocyte antigen (HLA)-A2 and HLA-DR1—augmented in vivo cytotoxic HLA-A2-restricted immune responses. Splenocytes from immunized mice were able to lyse syngeneic human prostate cancer cells; an effect that was absent from controls. Encouragingly, this immune response was accompanied by decreased tumour burden and increased overall survival.
