Acarbose is first hydrolyzed to +1 subsite (17.1 kcal/mol), then removed to −1 subsite (19.7 kcal/mol), and the second hydrolysis reaction is a rate-determining step. Three key crystal structures were obtained. The energy barrier of acarbose direct hydrolysis to M2 ( + 1/−1 subsites) is too high.
