ad, U2OS cells transfected with the ColorfulCell plasmid were incubated with LysoTracker Yellow and imaged live on an SDCM equipped with the multispectral camera system. The eight individual camera views (a) were acquired simultaneously, then processed by RLSU unmixing using a mixing matrix generated from the emission spectra of seven fluorophores (b) to generate unmixed images for the seven fluorophores (c).
