Phosphoinositide signal for lysosomal membrane repair

Maintenance of lysosome membrane integrity is crucial for cell homeostasis, and lysosomal membrane permeabilization (LMP) is a hallmark of lysosome-related diseases. Tan and Finkel now uncover a novel phosphoinositide-initiated membrane tethering and lipid transport (PITT) pathway required to rapidly resolve LMP associated with lysosome damage.

The authors found that phosphatidylinositol-4 kinase type 2a accumulates on damaged lysosomes resulting in local generation of the phosphatidylinositol-4-phosphate (PtdIns4P). PtdIns4P functions as a lipid messenger and recruits members of the oxysterol-binding proteins (OSBP)-related protein (ORP) family, most notably ORP9-11, to the lysosome.
