(A) Surface pre-TCR expression levels detected on SupT1 and JR.pTα pre-TCR+ cell lines by flow cytometry, using anti-CD3ε in combination with either drug-free anti-pTα monoclonal antibody, or anti-pTα-DM1 or anti-pTα-MMAE ADCs plus biotin-coupled anti-mouse (Fab’)2 plus PECy7-coupled streptavidin. Pre-TCR-negative Jurkat (TCRαβ+) and mock-transduced JR.MigR1 (TCRα) cell lines were assayed as negative controls. (B) Pre-TCR internalization in SupT1 cells pulsed with drug-free anti-pTα monoclonal antibody or anti-pTα ADCs at 4 °C for 30 min and incubated for the indicated times at 37 °C. Results are shown as mean percentages ±s.e.m. of pre-TCR+ cells detected by flow cytometry with a secondary anti-mouse IgG-FITC antibody normalized to time 0, (n = 3).
