Each bar represents a peptic peptide of Pfs48/45 D1D2 (ordered from N- to C-terminus based on the first amino acid of each peptic peptides). The deuterium uptake differences for each peptide at different time points are shown in orange (10 s), red (100 s), blue (1000 s) and black (10,000 s). The height of the vertical gray bar represents the sum of the mass differences for each peptide between the two states at each time point. The gray shades represent the differential uptake errors for each peptide, and the dashed horizontal lines represent the threshold which the uptake differences between the two states are considered significant if exceeded. This threshold corresponds to the 95% confidence interval, calculated from the pooled standard deviations for all peptides across all time points which is ±0.3 Da. The red and blue boxes highlight the regions with decreased and increased deuterium uptake in the bound state, respectively.
