BBP59 is not a basal body protein in bloodstream T. brucei. (A) BBP59 was endogenously tagged at its C-terminus with either a 3 × HA epitope (upper row) or a “Spaghetti monster”(SpMon)-Myc tag (lower row). Trypanosomes were fixed and labeled with an anti-Myc or anti-HA primary antibody followed by fluorescent secondary antibody, then stained with DAPI and analyzed by fluorescence microscopy. Trypanosome cell bodies are outlined in the “Merge” column. Scale bar = 20 μm. (B) Trypanosomes containing both V5-CK1.2 and BBP59-Myc epitope tags were labeled with anti-V5 and anti-Myc antibodies.
