The cells were transfected with miR-27b-3p inhibitor (HSC3) and mimic (HSC3-M3). The invasion (G, HSC3 and HSC3-M3) and wound healing (H, HSC3 and HSC3-M3) assays of cells after transfection showing the effect of miR-27b-3p on migration and invasion of the HNSC cell lines. Quantification results of invasion assay are additionally presented as bar graphs showing the relative invasion compared to the negative control group. The expression of (I, HSC3 and HSC3-M3) miR-27b-3p, (J, HSC3 and HSC3-M3) FN1 mRNA and (K, HSC3 and HSC3-M3) FN1 protein were shown to be related to the transfected with negative control sequence group, based on miR-27b-3p expression level regulation. The protein band intensity scanned and statistically analyzed by ImageJ. The relative expression of miRNA and mRNA were measured by RT-qPCR and the protein expression levels were analyzed by Western blot. Data are presented as the mean ± SEM of at least three independent experiments (N.S; non-significant).
