Nuclear factor (erythroid-derived 2)-like 2 (Nrf2) is a transcription factor that regulates the expression of proteins with various intracellular functions, however, the ligands that directly control the Nrf2 promoter are underexplored. Here, the authors identify small molecule ligands that regulate the expression of Nrf2 by targeting a newly identified i-motif-forming sequence in the Nrf2 promoter.
