Type II natural killer T cells use features of both innate-like and conventional T cells to recognize sulfatide self antigens

Natural killer T cells (NKT cells) recognize lipid antigens presented by CD1d. Zajonc and Rossjohn and their colleagues describe molecular interactions between type II NKT cell antigen receptors and CD1d-ligand complexes, which demonstrate distinct modes of recognition used by the receptors.

Invariant natural killer T cells recognize glycolipids from pathogenic Gram-positive bacteria

Natural killer T cells recognize glycolipid ligands in the context of the antigen-presenting molecule CD1d. Kronenberg and colleagues show that these cells recognize ligands derived from the pathogens Streptococcus pneumonia and group B Streptococcus.

A third population of human T lymphocytes that express αβ T cell antigen receptors with restricted α-chain diversity has been identified. These cells recognize the lipid glucose monomycolate from Mycobacterium tuberculosis presented by CD1b.

Control of CD1d-restricted antigen presentation and inflammation by sphingomyelin

Niemann–Pick disease is characterized by the cellular accumulation of sphingomyelin. Blumberg and colleagues use both mouse models and materials from patients with Niemann–Pick disease to show that sphingomyelin accumulation inhibits CD1d-restricted NKT cell activation and development.

A ligand-specific blockade of the integrin Mac-1 selectively targets pathologic inflammation while maintaining protective host-defense

Integrin-based therapeutics could block inflammatory processes but they also impair host defence, limiting their usefulness. Here the authors report an anti-Mac1 antibody that blocks its interaction with pro-inflammatory ligand CD40L but not other ligands, and show that it can protect against sepsis in mice.
