Mixed responses to targeted therapy driven by chromosomal instability through p53 dysfunction and genome doubling

Mixed responses to targeted therapy within a patient are a clinical challenge. Here the authors show that TP53 loss-of-function cooperates with whole genome doubling which increases chromosomal instability. This leads to greater cellular diversity and multiple routes of resistance, which in turn promotes mixed responses to treatment.

The metabolic dependencies of androgen receptor (AR)-driven growth in prostate adenocarcinoma are largely unknown but could represent a therapeutic target when hormonal manipulations fail. Here the authors demonstrate that the mitochondrial pyruvate carrier (MPC) is transcriptionally regulated by AR and that MPC inhibition suppresses tumour growth in hormone-responsive and castrate-resistant conditions.

This Review highlights the shortcomings in the evidence derived from such studies. M31’s massive and metal-rich stellar halo appears to indicate that a single dominant merger with a large galaxy took place about 2 Gyr ago, co-temporal with M31’s global burst of star formation. M32 is likely to be the stripped core of the disrupted galaxy.
