CircCDC42-encoded CDC42-165aa regulates macrophage pyroptosis in Klebsiella pneumoniae infection through Pyrin inflammasome activation

The function of circular RNA (circRNA) in the innate immune system is not fully understood. Here the authors characterise a cell division cycle 42 (CDC42)-165aa protein encoded by circRNA circCDC42 that promotes Pyrin-inflammasome mediated pyroptosis in Klebsiella pneumoniae (KP) infection and inhibition of circCDC42 reduces KP induced lung injury in mice.

Genome-wide CRISPR screens identify ILF3 as a mediator of mTORC1-dependent amino acid sensing

Yan et al. harness genome-wide CRISPR screens and identify ILF3 as a signalling intermediate negatively regulating mTORC1 activation upon amino acid sensing. ILF3 recruits GATOR2 to tether the GATOR complexes to lysosomes and regulate mTORC1 activity.

Mitochondrial fission induces immunoescape in solid tumors through decreasing MHC-I surface expression

Cancer cells downregulate surface expression of major histocompatibility complex I (MHC-I) for immune evasion. Here, the authors show that rapid mitochondrial fission activates the ER-stress response leading to reduced MHC-I complex formation and cell surface expression in solid cancer cells; moreover inhibition of mitochondrial fission increases the immune-mediated anticancer response in murine models.

A defined and simplified culture system for the derivation and growth of human induced pluripotent stem cells is reported. It permits increased efficiency of human reprogramming with an episomal approach. Also in this issue, Okita et al. describe methods for more efficient episomal reprogramming of human cells.

Snail1-dependent control of embryonic stem cell pluripotency and lineage commitment

Factors inducing epithelial to mesenchymal transition, such as the transcriptional repressor Snail1, have been implicated in cancer stem cell development and function. Here Lin et al.report that endogenous Snail1 is not required for embryonic stem cell pluripotency and self-renewal, but rather regulates Wnt-induced epiblast differentiation.
