  • Vpr is a HIV-1 accessory virulence factor that also interacts with the human DNA repair protein hHR23A. Here, the authors present the structure of Vpr in complex with the C-terminal half of hHR23A comprising the XPC-binding and ubiquitin-associated domains, which reveals that hHR23A interacts with the DCAF1-binding and not the substrate-binding Vpr surface and further illustrates how Vpr acts as a versatile structural adapter that targets diverse DNA repair pathways.

  • Mdm2 negatively regulates tumor suppressor p53 via binding to the transactivation and the DNA-binding domains of p53. Now work reveals a novel interaction between the N-terminal domain of Mdm2 and the extreme C-terminal region of p53, a region that bears the sites of many post-translational modifications that inhibit the Mdm2-p53 interaction.

  • The huntingtin protein (HTT) contains a polyQ tract preceded by an N-terminal flanking sequence (HTTNT) that contributes to HTT aggregation. Now the role of HTTNT in aggregation is explored in vitro, revealing a complex, multistep pathway initiated when polyQ disrupts HTTNT structure, enhancing the latter's assembly into prefibrillar aggregates. Within these intermediates, subsequent interactions of the polyQ moieties drive further assembly into compact amyloid aggregates.
