High resolution profiling of cell cycle-dependent protein and phosphorylation abundance changes in non-transformed cells

The cell cycle governs a precise series of molecular events, regulated by coordinated changes in protein and phosphorylation abundance, that culminates in the generation of two daughter cells. Here, the authors present a proteomic and phosphoproteomic analysis of the human cell cycle in nontransformed cells using deep quantitative mass spectrometry by isobaric labelling.

Conditional control of fluorescent protein degradation by an auxin-dependent nanobody

DNA damage during S-phase mediates the proliferation-quiescence decision in the subsequent G1 via p21 expression

Cell cycle arrest after DNA damage is achieved by the expression of the CDK inhibitor p21. Here the authors show that spontaneous DNA damage incurred in unperturbed cell cycles, leads to cell populations exhibiting a bistable state, with p53 and p21 regulating the proliferation-quiescence decision.

APC15 drives the turnover of MCC-CDC20 to make the spindle assembly checkpoint responsive to kinetochore attachment

The spindle assembly checkpoint halts cell-cycle progression in the presence of unattached kinetochores by preventing activation of APC/C. Pines and colleagues find that APC15 has a critical role in regulating APC/C activation by promoting release of the inhibitory MCC complex from APC/C once the spindle assembly checkpoint is satisfied.

UBE2S elongates ubiquitin chains on APC/C substrates to promote mitotic exit

The anaphase-promoting complex (APC/C), a ubiquitin ligase regulating mitotic progression, is a target for spindle assembly checkpoint. UBE2S, an ubiquitin-conjugating enzyme, is identified as a novel factor that elongates ubiquitin chains and promotes APC/C substrate degradation following release from the spindle assembly checkpoint.
