Ionizable lipids are essential components of lipid nanoparticles (LNPs) for efficient mRNA delivery, however, efficiently optimizing their chemical structures remains challenging. Here, the authors report a high-throughput platform for screening ionizable lipids using a STAAR approach, allowing for the synthesis and in vivo screening of novel lipids followed by a structure-activity study, leading to the development of CP-LC-0729, which along with its counterpart is capable of targeting the lungs.
