Carbene footprinting accurately maps binding sites in protein–ligand and protein–protein interactions

Mapping protein-ligand interactions is fundamental to advance the understanding of cellular processes and to develop drug discovery strategies. Here, the authors present a photo-activated probe that allows highly efficient labelling and identification of protein binding sites using mass spectrometry.

Concordant inter-laboratory derived concentrations of ceramides in human plasma reference materials via authentic standards

Here, the authors compared measurements between 34 laboratories from 19 countries, to quantify by mass spectrometry four ceramides of clinical relevance in human blood plasma Standard Reference Materials. The main goals were to evaluate concordance obtained in a large inter-laboratory trial and to report absolute concentrations of four circulating lipids in a publicly available standard.

Actin dynamics drive cell-like membrane deformation

The same type of polymer network deforms cell membranes inward, to absorb external material, and outward, to facilitate signal transmission. Experiments and theory show that these deformations are regulated by membrane tension and network mesh size.

A balance between membrane elasticity and polymerization energy sets the shape of spherical clathrin coats

A relationship between membrane tension and clathrin polymerization during endocytosis has not been experimentally established. Here, the authors show using an in vitroreconstituted system and theoretical modelling that membrane tension regulates clathrin polymerization into spherical cages by varying the membrane budding energy.

IRSp53 senses negative membrane curvature and phase separates along membrane tubules

The inverted-BAR domain protein IRSp53 associates with the inner leaflet of tubular membranes such as filopodia. Here, Prévostet al. demonstrate that the I-BAR domain of IRSp53 senses negative membrane curvature, and undergoes phase separation which may aid its clustering upon filopodia generation.
