NOTCH1 activation compensates BRCA1 deficiency and promotes triple-negative breast cancer formation

BRCA1 mutation carriers have higher chances of developing triple-negative breast cancer (TNBC). Here, the authors use the Sleeping Beauty mutagenesis system in Brca1 deficient mice and identify 169 putative driver genes, of which NOTCH1 accelerates TNBC formation through promoting epithelial-mesenchymal transition and cell cycle progression.

Synthetic lethality of RB1 and aurora A is driven by stathmin-mediated disruption of microtubule dynamics

Retinoblastoma susceptibility gene (RB1) is frequently mutated in lung cancers. Here the authors perform chemical and genetic vulnerability screens and identify aurora A kinase (AURKA) as a synthetic lethal candidate for RB1-deficient lung cancer cells and that AURKA inhibition sensitizes these cells to mitotic cell death.

Single-cell whole-exome sequencing is a powerful method for deciphering intratumor heterogeneity and identifying cancer drivers. Co-option of an extracellular protease for transcriptional control of nutrient degradation in the fungus Aspergillus nidulans

Li et al. report the PnmB protease as a moonlighting protein in the fungus Aspergillus nidulans as both a secreted protease that degrades extracellular nutrients and an intracellular protease that controls the turnover of the co-repressor NmrA. These results reveal a regulatory mechanism of the drivers of the nitrogen starvation response, as well as the functional evolution of a protease gene for transcriptional regulation and extracellular nutrient breakdown.
