The structure of human prostaglandin E receptor EP4 in complex with antagonist ONO-AE3-208 and a functional antibody reveals a ligand-binding site at the interface of the lipid bilayer that is unique among GPCRs.

Lysosomal lipid peroxidation contributes to ferroptosis induction via lysosomal membrane permeabilization

The early events in ferroptosis are not clear. Here, the authors report that intra-lysosomal lipid peroxidation (LPO) induces lysosomal membrane permeabilization (LMP) and consequent iron leakage, fostering cell-wide LPO and eventually ferroptotic cell death in tumor cells.

Inhibition of 7-dehydrocholesterol reductase prevents hepatic ferroptosis under an active state of sterol synthesis

Ferroptosis has been connected to liver disease through unclear mechanisms. Here, the authors identify the terminal enzyme of cholesterol synthesis, 7-dehydrocholesterol reductase, as a regulator of ferroptosis in hepatocytes that suppresses ferroptosis through 7-dehydrocholesterol accumulation.
