Investigating the neuronal role of the proteasomal ATPase subunit gene PSMC5 in neurodevelopmental proteasomopathies

Variants in the PSMC5 gene impair proteasome function and cellular homeostasis, altering brain development in children. This study reveals underlying molecular mechanisms contributing to this neurodevelopmental phenotype, and suggests therapeutic leads for neurodevelopmental proteasomopathies.

Suppression of gut colonization by multidrug-resistant Escherichia coli clinical isolates through cooperative niche exclusion

In this work, authors explore the potential of strain-specific commensal E. coli in their ability to inhibit multi-drug resistant Enterobacterales via nutrient competition. Their findings highlight the potential of rationally designed metabolically complementary probiotics for targeted gut decolonization of antibiotic-resistant bacteria.

Klebsiella oxytoca facilitates microbiome recovery via antibiotic degradation and restores colonization resistance in a diet-dependent manner

The authors demonstrate in a mouse model that Western-style diets synergize with antibiotics to impair microbiota function. In turn, Klebsiella oxytoca promotes post-antibiotic microbiota recovery and removal of the pathogen Klebsiella pneumoniae.

Functional characterization of a multi-cancer risk locus on chr5p15.33 reveals regulation of TERT by ZNF148

Genetic variants at multiple loci of chr5p15.33 have been associated with susceptibility to numerous cancers. Here the authors show that the association of one of these loci may be explained by a variant, rs36115365, influencing telomerase reverse transcriptase (TERT) expression via ZNF148.

A genome-wide association study identifies pancreatic cancer susceptibility loci on chromosomes 13q22.1, 1q32.1 and 5p15.33

Stephen Chanock and colleagues identify three new susceptibility loci for pancreatic cancer on chromosomes 13q22.1, 1q32.1 and 5p15.33. The association signal at 13q22.1 maps to a large nongenic region, whereas the signals at 1q32.1 and 5p15.33 map near the NR5A2 gene and CPTM1L-TERT region, respectively.
