Electron-density-informed effective and reliable de novo molecular design and optimization with ED2Mol

A deep generative model is developed for de novo molecular design and optimization by leveraging electron density. Wet-laboratory assays validated its reliability to generate diverse bioactive molecules—orthosteric and allosteric, inhibitors and activators.

Spectroscopic visualization and phase manipulation of chiral charge density waves in 1T-TaS₂

Two-dimensional charge density waves in layered semiconductors may exhibit chirality. Here, the authors utilize thermal annealing to reversibly switch the in-plane chirality of charge density waves in 1T-TaS₂ and demonstrate a vertical chirality-locking effect between the van der Waals-stacked layers.

An allosteric PRC2 inhibitor targeting the H3K27me3 binding pocket of EED

H3K27me3 binding to the EED pocket of the Polycomb repressive complex 2 (PRC2) is required to activate PRC2. An allosteric small-molecule inhibitor of PRC2 was identified that binds to the EED pocket and blocks PRC2 methyltransferase activity in cells.
