Noninvasive cardiac modulation via triplet-sensitized photoswitching in the phototherapeutic window

Azobenzene-based photoswitchable drugs traditionally require activation with UV-blue light having poor tissue penetration. Here, the authors show triplet sensitized photoisomerization of an azo drug with far-red and NIR light, allowing deep tissue penetration and in vivo heart rate control in frog tadpoles.

A robust molecular platform for non-volatile memory devices with optical and magnetic responses

The creation of ‘smart surfaces’ that can act as switches or memory devices will rely on systems with bistable states that can be interconverted externally. Now, a low-voltage and robust surface-confined switch that transduces an electrical input into an optical and magnetic output is described.

An allosteric modulator to control endogenous G protein-coupled receptors with light

Alloswitch-1 is a photoswitchable modulator for mGlu5, and it is the first photoswitchable allosteric GPCR modulator. It was generated by adding the azobenzene Ar-N=N-Ar scaffold into an existing positive allosteric modulator of the receptor.

Biological activity can be photoswitched by light-regulated drugs, but so far only diffusible ligands have been shown to work on endogenous receptors. Here the authors develop targeted covalent photoswitches that couple to a protein target by ligand affinity, and demonstrate photocontrol of GluK1-expressing neurons.
