Regulatory T cells aid stem-cell therapy for Parkinson’s disease

Inflammation caused by surgical trauma limits the survival of transplanted stem-cell-derived neurons in rodent models of Parkinson’s disease. Co-transplanting immune cells called regulatory T cells improves the therapy’s efficacy.

Approaching a cure for type 1 diabetes

A study has demonstrated that transplanting human embryonic stem cell–derived, insulin-producing cells shielded in capsules made with an optimized biomaterial can achieve long-term cure of diabetes in mice without the need for any immunosuppression.

An application-specific image processing array based on WSe₂ transistors with electrically switchable logic functions

Reducing circuit redundancy represents a priority for the scalability of parallel computing hardware. Here, the authors report the realization of pixel processing units consisting of single 2D WSe₂ transistors implementing electrically-switchable logic functions. This strategy enables the fabrication of an image processing array with ~16% transistor consumption compared to traditional circuits.

Feasibility of an implantable bioreactor for renal cell therapy using silicon nanopore membranes

An implantable bioartificial kidney with a cell-containing bioreactor could be used to treat end-stage renal disease. Here the authors demonstrate the feasibility of an implantable bioreactor by maintaining human cell viability and functionality after implantation in a xenograft model.

Loss of integrin α_vβ₈ on dendritic cells causes autoimmunity and colitis in mice

It is shown that mice deficient for β₈ integrin develop colitis, and that β₈ needs to be expressed by dendritic cells rather than T cells for colitis to develop. In addition, a decrease of T cells in the gut but not adjacent lymph nodes in diseased animals is shown. It is concluded that αVβ8-mediated integrin activation of dendritic cells is essential for prevention of colitis-inducing immune dysfunction.

Sun et al. identify the deubiquitinase USP15 as a regulator of PARP1 stability in triple-negative breast cancer and show that the hormone receptors ER, PR and HER2 inhibit USP15-mediated PARP1 stabilization, modulating base excision repair.

Islet encapsulation therapy — racing towards the finish line?

Transplanting pancreatic islets in immune-isolating capsules might cure type 1 diabetes mellitus while avoiding the immunosuppression that is normally needed to protect transplanted islets from rejection. A recent study demonstrates that allogeneic islets can survive in capsules with improved biocompatibility without immunosuppression in non-human primates, bringing us one step closer to applying this therapy in humans.

The inhibitory protein PD-1 is expressed on activated T cells. Fife and colleagues find that interactions between PD-1 and its ligand PD-1L are needed to maintain tolerance and prevent interactions between tolerized T cells and dendritic cells.
