Somatostatin neurons in prefrontal cortex initiate sleep-preparatory behavior and sleep via the preoptic and lateral hypothalamus

The sleep drive builds with time spent awake. Tossell et al. show in mice that tiredness triggers neurons in the prefrontal cortex to instruct the hypothalamus to initiate nesting and then sleep itself, ensuring that sleep occurs in a safe place.

GABA and glutamate neurons in the VTA regulate sleep and wakefulness

Yu, Li et al. show that VTA GABA and glutamate neurons induce sleep and waking, respectively, via projections to the lateral hypothalamus and nucleus accumbens. Thus, in addition to influencing reward-directed behaviors, the VTA regulates arousal.

Neuronal ensembles sufficient for recovery sleep and the sedative actions of α₂ adrenergic agonists

The authors use TetTag pharmacogenetics to mark neuronal ensembles activated in the preoptic hypothalamus during dexmedeotomidine-induced sedation or recovery sleep. When these ensembles were selectively reactivated, NREM sleep and the accompanying drop in body temperature were recapitulated. Thus α₂ adrenergic receptor–induced sedation and recovery sleep share circuitry sufficient for producing these states.
