The enterococcal cytolysin is a two-component toxin that is responsible for a fatal form of alcoholic hepatitis. Here, authors used knowledge of the toxin’s biosynthesis to develop small molecule inhibitors of toxin production.

Irreversible electroporation augments checkpoint immunotherapy in prostate cancer and promotes tumor antigen-specific tissue-resident memory CD8+ T cells

Irreversible electroporation (IRE), a soft-tissue ablation technique used for tumour ablation, has been suggested to promote systemic immune responses. Here the authors show that IRE, followed by anti-CTLA-4 blockade, elicits the expansion of tumor antigen-specific CD8+ T cells and is associated with tissue residency and improved anti-tumor immune response in a preclinical model of prostate cancer.

An unusual carbon–carbon bond cleavage reaction during phosphinothricin biosynthesis

The biosynthesis of the naturally occurring compound phosphinothricin tripeptide (PTT) involves the conversion of 2-hydroxyethylphosphonate (HEP) to hydroxymethylphosphonate (HMP). This requires the cleavage of a C(sp³)–C(sp³) bond. Here, the X-ray crystal structure of the enzyme that catalyzes this reaction (hydroxyethylphosphonate dioxygenase, HEPD) is solved, and the mechanism of this remarkable transformation is explored.
