Noise amplification and ill-convergence of Richardson-Lucy deconvolution

Computer algorithms for contrast and resolution enhancement of microscopic images can become derailed. The authors here provide proof is offered that the Richardson-Lucy algorithm turns out to amplify noise in the initial data, wrecking the final outcome.

Multispectral live-cell imaging with uncompromised spatiotemporal resolution

A tree-like arrangement of dichroic mirrors and multiple cameras coupled with an iterative spectral unmixing algorithm enables multispectral imaging of live cells in up to eight spectral channels with diffraction-limited spatial resolution and temporal resolution of 0.3 s for imaging a full cell volume.

Simultaneous orientation and 3D localization microscopy with a Vortex point spread function

Molecular orientation is often ignored during single-molecule localisation microscopy. Here, the authors use a Vortex point spread function in order to simultaneously estimate the 3D position, dipole orientation and degree of rotational constraint, within 30% of the Cramér-Rao bound limit.

Measuring image resolution in optical nanoscopy

A method for computing the intrinsic resolution of a super-resolution image that accounts for localization uncertainty, labeling density and image anisotropy is described. This work extends and builds on the Fourier ring correlation method used in cryoelectron microscopy.
