Esterase mutation is a mechanism of resistance to antimalarial compounds

Pepstatin is a known inhibitor of malarial proteases, but its activity varies between sources. Here, Istvanet al. identify a pepstatin ester as the active component of pepstatin preparations and show that this prodrug is activated by a Plasmodiumesterase, mutation of which can confer resistance to pepstatin and other compounds.

Unbiased screening identifies regulators of cell-cell adhesion and treatment options in pemphigus

Cell-cell adhesion by desmosomes is weakened in the blistering skin disease pemphigus vulgaris. Here, authors found that the transcription factor Kruppel-like factor 5 is essential for the expression of the desmosomal gene desmoglein 3 and suggest a novel treatment approach.

Quantitative measurement of antibiotic resistance in Mycobacterium tuberculosis reveals genetic determinants of resistance and susceptibility in a target gene approach

Molecular diagnostics for tuberculosis have focused on predicting drug susceptibilities in a binary manner (i.e., strains are either susceptible or resistant). Here, CRyPTIC Consortium researchers use whole genome sequencing and a quantitative assay to identify associations between genomic mutations and minimum inhibitory concentrations in over 15,000 Mycobacterium tuberculosis clinical isolates.
