Structural and mechanistic insights into azetidine-associated αKG-NHFe enzyme OkaE with multifunctional catalysis

OkaE is an αKG-NHFe enzyme capable of synthesizing the pharmacophoric motif azetidine ring via unique C–C bond formation. Here, the authors report the multifunctionality of OkaE, which enables not only azetidine formation but also scaffold diversification through sequential oxidations, including hydroxylation, epoxidation, and ring cleavage.

Asgard Arf GTPases can act as membrane-associating molecular switches with the potential to function in organelle biogenesis

The formation of organelles was a pivotal point in eukaryotic evolution. Here they show that Archaea possess Arf-like GTPases that can perform key organelle-producing mechanisms when expressed in a eukaryotic cell, laying the foundation for the evolution of endomembrane organelles.

RETRACTED ARTICLE: Endoperoxide formation by an α-ketoglutarate-dependent mononuclear non-haem iron enzyme

The X-ray crystal structures of FtmOx1, the first known α-ketoglutarate-dependent mononuclear non-haem iron enzyme that can catalyse an endoperoxide formation reaction, are presented, along with further biochemical analyses which reveal the catalytic versatility of mononuclear non-haem iron enzymes, and help to unravel the mechanisms of endoperoxide biosyntheses.

Monoclonal antibody (mAb) therapy is now commonplace in the clinic, yet such reagents can elicit unwanted side effects due to interactions with Fcγ receptors. Georgiou and colleagues have engineered mAbs that lack such FcγR interactions but retain the ability to activate complement and show that these modified mAbs have efficacious effector function.
