Allowed Conformations of Alanyl preceding Prolyl Residues

Abstract

THE number of possible conformations of peptide sequences containing the bulky prolyl residue is limited because of steric restrictions. Schimmel and Flory1,2 have demonstrated that the “Ra” conformation (for explanation see ref. 3) in the alanine independent unit is disallowed for all amino-acids except glycine if they precede a prolyl residue. A prolyl–prolyl sequence would be totally disallowed, its energy being about 60 kcalories/mole above the deepest minimum. Leach et al.4 concluded that this Ra region is allowed not only for alanyl followed by prolyl, but also for prolyl followed by another prolyl.
