Effect of the supernatant of isolated cholangiocytes (after 6 h of incubation at 37°C) from 3-day BDL WT and KO mice on the proliferation of mouse cholangiocyte cell lines. The mouse cholangiocyte cell line was also pretreated in vitro with the CGRP receptor antagonist (CGRP8–37, 1 μM) for 30 min prior to the addition of the supernatant from 3-day BDL WT mice. After 24 h of incubation, cholangiocyte proliferation was measured by MTS proliferation assay. We found that supernatant from 3-day BDL WT mice stimulates the proliferation of cholangiocytes compared to basal proliferative rates. Consistent with our in vivo data, supernatant from BDL KO mice did not significantly stimulate proliferation. Indeed, the stimulation of cholangiocyte proliferation by BDL WT mouse supernatant was blocked by in vitro pretreatment with CGRP8–37. Data are mean±s.e.m. of seven experiments. *P<0.05 vs basal.
