A proposed model for p53/p21/mdm-2 regulation in germ cell tumors. A, Intratubular germ cell neoplasia (ITGCN): p53 induces p21 in the absence of mdm-2. p21 binds and inactivates cyclin/cdk complexes and inhibits their activity, resulting in cell cycle arrest. B, Invasive tumor: mdm-2 binds p53 and blocks induction of p21, allowing for cyclin/cdk activity and continued cellular proliferation.
