Effect of 15d-PGJ₂ on negative regulators of Stat3 signalling. (A) Effect of 15d-PGJ₂ on SOCS3 expression. SCC9 cells were either treated with 20 μ M, 15d-PGJ₂ (PG) or rIL-6 (25 ng ml⁻¹) for 1 h. Cells were blotted with SOCS3 (M-20) antibody and subsequently stripped and reprobed with actin control antibody. (B) Effect of 15d-PGJ₂ on SHP2 phosphorylation. SCC9 cells were treated with 20 μ M, 15d-PGJ₂ (PG) for 10 min. Cell lysates were immunoprecipitated with SHP2 antibody and Western blotted for phosphorylated (PY) and total SHP2.
