AKT phosphorylation determines the activation of NF-κB cascade and, in parallel, the phosphorylation and inhibition of BAD molecule. (a) AKT phosphorylation (Ser 473) was monitored in keratinocytes treated with IFN-γ/TNF-α at different time points by WB analysis. Filters were stripped and re-probed with an anti-AKT Ab. (b) Primary human keratinocytes were pre-treated with Ly294002 (15 μM) for 2 h and then stimulated with IFN-γ/TNF-α for 12 h. In left panel, protein extracts were subjected to WB with anti-phospho AKT (Ser473), anti-AKT, anti-IκBα, anti-phospho p65 (Ser 276), anti-BCL-2, anti-BCL-XL and anti-β-actin Abs. In right panels, co-immunoprecipitation of endogenous phospho-BAD (Ser136) was followed by WB using anti-BCL-XL Ab. Filter was stripped and re-probed with BAD Ab
