Pre-incubation with tubastatin A regulates the activity of peroxiredoxin 1. 661W cells were treated with tubastatin A (TST) for 8 h as indicated, or with H2O2 (200 μM) for 6 h, and they were preincubated with 1, 5 and 10 μM TST for 2 h followed by incubation with H2O2 for 6 h (TST+H2O2). (a) Immunoblot analysis of cell extracts revealed an H2O2 induced increase in the level of PrxSO2/3, the inactivated form of peroxiredoxin 1 (Prx1), which was significantly reduced by pre-incubation with 10 μM TST, as shown by quantitative evaluation of the immunoblot (b). ac Tub, acetylated α-tubulin; α-Tub, α-tubulin. n=4
