Quantification of replicated viral DNA 24 h after infection. HeLa cells (1 × 105) were infected with AdTRE-E1 and AdCMV-rtTA at multiplicity of infection (MOI) 10 and then incubated with (open square) or without (closed square) Dox for 24 h. Viral DNA was isolated from the cells and analyzed by the real-time PCR quantification method to evaluate adenoviral E1 and E4 copy numbers, as described in Materials and methods. E1 and E4 copy numbers were normalized by the β-actin DNA copy number. Mean±s.e. of triplicate determination is shown.
