Dialling up the circadian clock to target ageing

Targeting ageing remains challenging owing to its multifactorial nature and the interplay between genetic, metabolic and environmental drivers. Circadian rhythms have emerged as regulators of multiple ageing hallmarks, yet their therapeutic relevance remains underexplored. Now, in an article in Cell, Zhao et al. show that pharmacologically enhancing circadian amplitude in the hypothalamic paraventricular nucleus (PVN) can reverse ageing phenotypes and extend lifespan in aged male mice.
