(a) Immunoblotting of whole cell protein extracts from T84 (left panels) and Caco2 (right panels) epithelial cell monolayers show phosphorylation of STAT5 and STAT1 after IFNγ (10 ng/ml) exposure. (b) Different kinetics of STAT1 and STAT5 phosphorylation were induced by IFNγ in T84 cells and PMA-differentiated THP-1 monocytes. Bright-field micrographs ( × 20 magnification) below the immunoblots demonstrate the morphology of THP-1 cells before and following 48 h of treatment with 20 nM PMA. Src kinase inhibitor PP1 (125 nM) (c) or Fyn siRNA treatment (d) of T84 cells did not inhibit IFNγ-induced phosphorylation of either STAT5 or STAT1 (images are representative of three experiments).
